On June 16, 2015, Fluellen signed a contract with the Buffalo Bills. For final roster cuts, he was released on September 4, 2015, but re-signed with the team on September 7 and was cut from the team once again on September 11, 2015. He was then re-signed to the active roster the following day.

On October 7, 2015, Andre Fluellen was signed by the Detroit Lions after Tyrunn Walker was placed on injured reserve. it was the ninth time Fluellen had signed with the Lions. On December 9, 2015 Fluellen was released from the injured reserve list by the Lions, making him a free agent.

On April 9, 2013, Fluellen was signed by the Chicago Bears. Fluellen was released by the Bears on May 20, 2013.

On July 26, 2013, Fluellen was signed by the Lions, marking the third time in his career that he joined the team. After redshirting his initial year at Florida State, Fluellen appeared in all 12 games of the 2004 season. Serving as a back-up to Brodrick Bunkley, he finished the year with six tackles, including one for loss, and two pass breakups. Against both Miami (FL) and Duke he recorded two tackles each. In his sophomore season, Fluellen replaced Travis Johnson at defensive tackle. Recording 26 tackles on the year, he ranked second on the team behind Bunkley for most tackles by a defensive lineman. Fluellen also had 7.5 tackles for loss, and tied for third on the team in quarterback hurries with twelve. Season-highs in both tackles (eight) and tackles for loss (two) he recorded against North Carolina State.
